Description

Principle of the assay: human CD244 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CD244 has been precoated onto 96-well plates. Standards(NSO, C22-R221) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CD244 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human CD244 amount of sample captured in plate.
Background: CD244 (Cluster of Differentiation 244) is a human protein encoded by the CD244 gene. It is also known as Natural Killer Cell Receptor 2B4. Members of the CD2 family adhere to each other instead,the cell surface glycoprotein 2B4 is related to CD2 and is implicated in the regulation of NK- and T-cell function. The 2B4 gene was mapped to 1q22 on gene map, where SLAM, CD48, CD84 and LY9 are also located. Using recombinant human NK cell-activating ligand 2B4 fused to domains 3 and 4 of rodent Cd4 and flow cytometric analysis, It was found that CD48 binds to 2B4. BIAcore surface plasmon resonance analysis showed that the affinity of CD48 for 2B4 is approximately 10-fold higher than that shown for CD48 and CD2
